Perinatal elimination of genetically aberrant human cortical neurons

Abstract Human neurons are postmitotic and long-lived, requiring precise genomic regulation. Normal neuronal function is highly dosage-sensitive, with copy number variants (CNVs) and heterozygous point mutations associated with a host of neurodevelopmental and neuropsychiatric conditions 1–4 . Here we investigated somatic CNVs arising in single neurons to understand the dynamic processes that shape the normal neuronal genetic landscape. Altogether, we sequenced 2,357 single neurons from human postmortem brain from fetal to postnatal life. Two single-cell whole-genome sequencing approaches—Tn5-transposase-based amplification (1,526 neurons, 16 individuals) and primary template amplification 5 (831 neurons, 12 individuals)—showed that an average of 25.9% [12.7–46%] of prenatal cortical neurons have highly aberrant genomes, that is, widespread CNVs involving multiple chromosomes. This proportion reduced by about 4.5-fold after birth across the two single-cell whole-genome sequencing approaches (Fisher meta-analysis P = 3.5 × 10 − 6 ). Neurons with widespread CNVs were mainly eliminated perinatally, timed roughly parallel to periods of programmed cell death 6 , whereas neurons with smaller CNV burdens declined slowly in frequency with postnatal age. CNVs in surviving neurons were depleted for dosage-sensitive genes and genes involved in neurodevelopmental disorders (hypergeometric test P < 0.05), suggesting selective elimination of neurons with these CNVs. Analysis of 44,861 nuclei with single-nucleus assay for transposase-accessible chromatin (snATAC) and single-nucleus RNA sequencing (snRNA-seq) showed that neurons with high CNV burdens exhibited abnormal synaptic gene expression, suggesting that abnormal synaptic gene regulation contributes to neuronal elimination. Elimination of defective neuronal genomes might represent an important developmental genome quality-control process that may be vulnerable in neurodevelopmental disease, and suggests purifying selection during human neurodevelopment.

Mark A. Ratner (1942-2026): Founded molecular electronics while spearheading the growth of science at Northwestern University.

Recently, we lost one of the original developers of the field of molecular electronics. Mark A. Ratner wrote a famous paper in 1974 that developed the theoretical framework for making a molecule act as a rectifier. This idea, and a host of further ideas that were stimulated by this and other papers, led to a huge amount of experimental and theoretical studies that continue to this day. But Mark's career was much more than that, with leadership at Northwestern University and beyond that continues

Historical trends show more declining than increasing species among European pollinators.

Anthropogenic global change presents a growing threat to insect pollinators. Yet, long-term, continental-scale assessments of their population trends remain limited. Here, we leveraged a curated database of over four million occurrence records, covering 88% of extant bee and hoverfly species in Europe, to assess persistence, richness, and occupancy trends over the past century. Species persistence at the European level was relatively high, with modern records available for 87% of bee and 89% of

The ABCD hypothesis.

If [Formula: see text] and [Formula: see text] are cardinals, then the cardinal exponentiation [Formula: see text] is the set of all functions from [Formula: see text] into [Formula: see text]. The ABCD hypothesis is the statement that for all infinite cardinals [Formula: see text] and [Formula: see text], [Formula: see text] if and only if [Formula: see text] and [Formula: see text]. The Axiom of Determinacy, AD, states that every two-player infinite game on [Formula: see text] has a winning st

The delocalized nature of chemical bonding at a catalytic metal surface.

Adsorbate bonding at metal surfaces requires hybridization of localized molecular orbitals with delocalized electronic bands of the metal; hence, the electronic wave function involved in bonding may extend far beyond the adsorption site. We revealed the consequence of this delocalization by determining the distance over which structural variations of the metal surface influence the molecule-metal bond strength. Velocity-resolved kinetics provided adsorption energies for three molecules bound at

Chiral altermagnon in MnTe.

Altermagnetism has been recognized as a magnetic phase that bridges the properties of ferro- and antiferromagnetism. The momentum-dependent spin splitting observed in these materials reflects their distinctive symmetry characteristics, which also establish the conditions for chiral magnons to emerge. Here we provide direct experimental evidence for a chiral magnon in the altermagnetic candidate manganese telluride (MnTe) revealed by circular-dichroism resonant inelastic x-ray scattering. This mo
